Sinusoidal cells make up about onethird of the total number of cells in the liver. This cell group forms a coordinated defense system designed to protect the liver from injury. The most potent of the parenchmal macrophages, the Kupffer cell, performs a variety of functions necessary for normal liver operations.

It has been over 100 years since the discovery of phagocytic mesenchymal cells in the lining of the hepatic sinusoids (2:495). Because of their ability to take up vital dyes, the sinusoidal cells of the liver were grouped together with reticular cells, histiocytes, and monocytes. Collectively these were called the reticuloendothelial system (2:495).

Two distinct cell types line the hepatic sinusoids, one of which is the Kupffer cell. Kupffer cells are distinguished in rat liver by the positive peroxidase reaction in their endoplasmic reticulum and nuclear envelope (2:495).

In general, Kupffer cells have the morphological characteristics typical of macrophages (3:508). Their cytoplasm is bulky and contains abundant, welldeveloped lysosomes and pintocytotic vesicles. Indeed, the volumetric composition of Kupffer cells is 13.5% lysosome and 2% pinocytotic vesicle. They also tend to be stellate in shape and of relatively large diameter (8.7 to 9.1 micrometers). When observed by scanning electron microscope, Kupffer cells also exhibit surface structures such as lamellopodia and microvilli.
